Welcome to Fluorescent Microscope
Fluorescent microscopy is the science of using specialized optical equipment to produce a specimen image using light that fluoresces from the specimen itself. The fluorescent microscope uses a light source such as mercury or xenon arc lamps. These produce a high intensity light that is capable of causing fluorescent molecules to fluoresce. The light source is a shorter wavelength of light and is passed through a filter cube to get a narrow bandpass of wavelengths to be incident on the specimen. This is called the exciting (for excitation) filter.
The microscope’s dichroic filter cube then picks up the reflected and emitted light from the specimen and filters out all but the desired fluorescing wavelength which is of a longer wavelength and very faint. This filter cube is called the emission filter and is also a narrow bandpass filter that has been carefully selected to match the biological specimen’s natural or stained fluorescent
wavelength. Some biological specimens naturally fluoresce, such as chlorophyll. Other samples need to be tagged by treating them with a chemical to cause them to fluoresce.
The fluorescent microscope is a much desired microscopy tool for medical and biological research as it allows imaging and identification of particulars cells and cellular components. The particular fluorescent dye used for example may allow the researcher to distinguish between live or dead cells, or cells with cancer. Green fluorescent protein is used by researchers in live cell imaging under the fluorescence microscope. Researchers from a wide array of discipline areas use this, such as in pharmacology, neurobiology, microbiology, cell biology studies, biomedical research, and environmental control.
